The protein encoded by this gene is a member of the TNF-receptor superfamily. This protein is one of the major receptors for the tumor necrosis factor-alpha. This receptor can activate NF-kappaB, mediate apoptosis, and function as a regulator of inflammation. Antiapoptotic protein BCL2-associated athanogene 4 (BAG4/SODD) and adaptor proteins TRADD and TRAF2 have been shown to interact with this receptor, and thus play regulatory roles in the signal transduction mediated by the receptor. Germline mutations of the extracellular domains of this receptor were found to be associated with the autosomal dominant periodic fever syndrome. The impaired receptor clearance is thought to be a mechanism of the disease.
Soluble TNF Receptor Inhibitor Human Recombinant produced in NSO is a disulfide linked homodimer, glycosylated, polypeptide chain having a molecular mass of 48Kda but as a result of the glycosilation, the recombinant protein migrates at 55-60Kda protein in SDS-PAGE.
sTNFRI /Fc chimera (sTNF RI/Fc chimera) was fused to carboxy-terminal 6X histidine-tagged Fc part of human IgG1, via a linker peptide.
sTNFRI is purified by proprietary chromatographic techniques.

It is recommended to reconstitute the lyophilized Soluble TNF Receptor Inhibitor Human Recombinant in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Lyophilized Soluble TNF Receptor Inhibitor although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution sTNFRI Human Recombinant should be stored at 4°C between 2-7 days and for future use below -18°C.

The sequence of the first five N-terminal amino acids was determined and was found to be Met-Glu-Glu-Val-Ser.
Greater than 95.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.
The activity was measured by its ability to neutralize apoptosis of mouse L929 cells treated with 0.25 ng/mL recombinant human TNF-a. The ED50 for this effect is typically 0.4-1 ng/mL.
